SLC2A1
SLC2A1全稱為葡萄糖轉(zhuǎn)運(yùn)蛋白1（GLUT1），是人體內(nèi)分布最廣的葡萄糖轉(zhuǎn)運(yùn)載體，在血腦屏障、紅細(xì)胞和胎盤等組織中高表達(dá)。SLC2A1通過促進(jìn)葡萄糖跨膜運(yùn)輸維持腦部能量供應(yīng)，對中樞神經(jīng)系統(tǒng)功能至關(guān)重要。GLUT1缺陷綜合征是其最典型的遺傳性疾病，患者因轉(zhuǎn)運(yùn)功能障礙出現(xiàn)嬰兒期癲癇、運(yùn)動障礙和智力發(fā)育遲緩，臨床通過腦脊液葡萄糖檢測結(jié)合基因篩查確診，生酮飲食替代供能是目前主要干預(yù)手段。膠質(zhì)母細(xì)胞瘤、乳腺癌等惡性腫瘤中GLUT1常異常高表達(dá)，與腫瘤代謝重編程密切相關(guān)，這使得SLC2A1成為抗癌藥物開發(fā)的重要靶點(diǎn)。例如小分子抑制劑BAY-876已在臨床前研究中顯示出阻斷腫瘤葡萄糖攝取的效果。研究人員正探索通過調(diào)控GLUT1活性改善糖尿病并發(fā)癥及阿爾茨海默病能量代謝障礙的新策略。

IHC image of CSB-RA297401A0HU diluted at 1:100 and staining in paraffin-embedded human cervical cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4℃ overnight. The primary is detected by a Goat anti-rabbit IgG polymer labeled by HRP and visualized using 0.05% DAB.
